Abstract
The fish eggs and larvae in genus Repomucenus are dominant in the coast of Yellow Sea , Korea. Morphologically, they can be easily distinguished from other family or genus. The eggs are characterized by a hexagonal egg membrane, no oil globule, and the small size ranged from about 0.6 to 0.85 mm. The larvae have a big head and three melanopore lines at the lateral body. However, their morphological similarities among related species within genus level make difficult to identify them into species level. For species identification of eggs and larvae in genus Repomucenus, DNA barcoding was used. The 16S partial gene of mtDNA was analyzed as the DNA barcode to classify the species. The DNA of the eggs and larvae were extracted using chelex-based method and amplified at the 16S gene. In the result of that, we can identify the eggs and larvae of Repomucenus as follows; R. koreanus, R. ornaptipinnis, R.curvicornis, R. valenciennei. The larvae of R. koreanus is the first record.
