Abstract
To study bacterial community associated with a marine sponge sample of tropical area of Micronesia, we performed T-RFLP (terminal-restriction fragment lemgth polymorphism) analysis in comparison with that of its surrounding sea water based on 16S rDNAs gene. Also, cloning and sequencing of bacterial 16S rRNAs were carried out for the characterization of each bacterial component with terminal restriction fragments (T-RFs). Total 14 of T-RF peaks generated from a restriction enzyme, HaeIII were detected in seawater. Among them, 9 T-RFs were identified through 16S rDNA clone analysis, the which major T-RF peak (46.8% of the total clone libraries) with 291/292 base was affiliated in the Cyanobacterial phylum. Total 16 T-RFs by tetrameric restriction enzyme HaeIII, HhaI and MspI, obtained in the marine sponge tissue, were identified through 16S rDNA clone analysis. Major T-RF peaks with 68, 98 and 160 base in length (73.1% of the total clone libraries) were affiliated within r-Proteobacteria.
