Abstract
The aims of this study were to identify and investigated the role of hydrogenases for better understanding of metabolism in Thermococcus onnurineus NA1, a sulfur-reducing hyperthermophilic archaea, which was isolated from a deep-sea hydrothermal vent area. We analyzed complete genome sequence and investigated the distribution and gene clusters of hydrogenases and related proteins. The genome analysis revealed an extraordinary proportion of hydrogenases and related proteins (5.5%) are present in a complete genome. Two membrane-bound hydrogenases (Mbh and Mbx) and two cytoplasmic NiFe-hydrogenases (Sulf-I and Sulf-II), reported in Pyrococcus spp. and T. kodakaraensis, are conserved. Three additional hydrogenase clusters (Hyg4-I, Hyg4-II, Hyg4-III) exhibited unique primary sequences and showed similarity to components of hydrogenase-4 from Rhodospirillum rubrum (a Gram-negative, purple-colored Proteobacterium) and P. abyssi. The Hyg4-III gene cluster was found to be associated with the a/b/g subunits of F420 hydrogenase, of which homologoues have not been reported from Thermococcales.
