Abstract
- To study the genetic diversity and population genetic structure of the fat greenling (Hexagrammos otakii), a total of one hundred and fifty five individuals from the yellow sea, the Southern sea of Korea, the East sea and the north western Pacific were investigated. Genetic variations in the mtDNA COI, COIII-ND3-ND4L and Cytb gene sequences were analyzed. The haplotype diversity ranges from 0.97 to 0.99% indicating a high level of genetic diversity. A total of 135 polymorphic sites were found, and 92 haplotypes were defined in the concatenated sequences of those gene regions. The demographic history examined by neutrality tests and mismatch distribution analysis indicated a Pleistocene population expansion at abou 125000-184000 years. The haplotype network showed a star burst structure suggested a very recent divergence of most haplotypes. Hierarchical molecular variance analysis(AMOVA) and conventional population FST comparisons revealed that no significant genetic structure was established throughout the examined range of distribution for the species. The knowledge on genetic diversity and genetic structure will be vital to establish appropriate fishery management strategy for the species.
