Abstract
A new species of Haematococcus sp. found in Korea was cultured in different media such as Hong Kong (HK), Fortified Bold’s Basal Medium (FBBM) and Affan-Kang-Haematococcus (AKH) media to select best growth media at the laboratory. Then, best medium was used for roof-based Haematococcus sp. culture which was carried out for three weeks in late autumn of 2010. The Biomass production, doubling time and biochemical composition was analyzed. During the roof-based study, temperature and pH varied from 18.20 to 31.50 °C and 7.50 to 9.50, respectively. The biomass production was 96±0.02, 140±0.04 and 285±0.04 mg/L in HK, FBBM and AKH media, respectively at laboratory, and the biomass production at roof was 141±0.04 mg/L which was similar with the biomass production of FBBM medium at laboratory culture. Doubling time was 0.14, 0.10 and 0.06 d-1 in HK, FBBM and AKH medium respectively, and in roof the doubling time was 0.14 d-1 which similar with that of HK medium. Biochemical composition such as protein, lipid, carbohydrate, ash and moisture of roof-based cultured Haematococcus sp. was 37.87±0.12, 13.49±0.61, 15.98±0.69, 25.61±0.08 and 7.05±0.26%, respectively. Further study of determination of astaxanthin and carotenoid is going on.
