Abstract
A few microorganisms have been identified as pathogenic agents responsible for various outbreaks of marine organism disease. But most of diseases have been implicated within a subset of marine organism in our knowledge of the host range and geographic extent of a given pathogen. In this study, we purposed the rapid and inexpensive detection of pathogens in coral, seaweed and sponge species using PCR-based assays. Total 5 pathogen-specific vibrio bacteria primer sets and 2 pathogen-specific 16s rDNA primer sets were designed for detecting coral, seaweed and sponge pathogenic disease. The pathogenic bacteria detected in coral were Aurantimonas coralicida, Serratia marcescens, Vibrio shilonii, Vibrio coralliilyticus, Vibrio carchariae, and vibrio bacteria were detected in seaweed and sponge. The assay was applied to coral samples from the Jeju Island, Japan and Taiwan, and to seaweed samples from Dokdo, Jeju, Yeosu and Uleongdo, and to sponge samples from Jeju in korea. After the identification of infected samples, the PCR-based assay is to be evaluated for detection of presence of pathogenic bacteria in coral, seaweed and sponge.d geographic extent of a given pathogen.
