Abstract
The rockfish Sebastes schlegeli is one of aquaculture species in Korea. Low temperature would affect physiology of the fish. The rockfish may exhibit reduced feeding and growth during winter time. To identify genes of S.schlegeli responding to the low temperature, we used the DDRT-PCR method as a means of analysing differentially expressed genes. From cold acclimated fish at 4℃ for 10 days, two up-regulated and one down-regulated cDNA were detected and sequenced. The full length sequence of three cDNA were obtained by the 5'-RACE PCR. Two up-regulated genes were turn out to be a gene (TubaSS: 432 a.a) coding for alpha-tubulin and an unknown protein gene (SSBRDEG16B: 64 a.a). One down-regulated cDNA was a gene (S100CBPSS: 84 a.a) coding for S100 like protein. The level of differential expression of the gene, TubaSS and S100CBPSS was examined by the real-time PCR methods. The gene expression patterns confirmed with the DDRT-PCR result. The relative expression level of TubaSS was 1.3 fold higher than that at 4℃. On the other hand the expression level of S100CBPSS was down-regulated at 4℃by 2.3 fold. In this study, we first identified a few genes of S.schlegeli which show differential expression in response to cold acclimation.
