Abstract
DNA chip technology is very developed tool for the gene analysis and diagnosis of illness. This method consists of probes that can detect variation of many genes and is very rapid, simple, and economic than the other established methods. We applied this method for skate species identification. Skates and rays (Order Rajiformes) are unique among Chondrichthyes for their high species diversity and morphological conservatism. Skates are commercially very important species in Korea and due to high consumer demand for these fishes, the amount of imported the fish from the world is increases. However, the method of discriminating between domestic and imported species of the skates has not established yet because of morphological similarities among the species and of high degree of variations. The confusion in systematics is even in the family-level classification. In this study, we aim to resolve the problems in species identification among the Korean skates and imported species by mitochondrial DNA barcode analysis and oligonucleotide DNA-chips. This study shows the results of mtDNA typing by oligonucleotide microarray were identical with that of sequencing analysis. Also, this method is very rapid, simple, and economic than the established mtDNA sequence typing method. Enormous hurdles must still be overcome, we confident that this method can be used as an alternative method for species identification.
