dc.description.abstract Pacific squid(Todarodes pacificus) is a popular and commercial species in Korea sea. Information on the diet of Todarodes pacificus is scarce due to morphologically recognizable remains in their digestive tracts. This study reports on the development and application of genetic toos for the identification of prey remains recovered from Pacific squid gut contents. And we explored the use of polymerase chain reaction(PCR)-based methods for detection of DNA in the prey remains and amorphous slurry from a pacific squid gut sample. The COI gene of Mitochondrial DNA amplified, allowing separation of prey components. And we found out Todarodes pacificus, Engraulis japonicus, Maurolicus mueller, Thysanoessa longipes, Amphipoda, Decapoda and Copepoda in the gut. -
