dc.description.abstract | To investigate the long term effects of artifitial DNA (plasmid) originated from LMO (living Modified Organisms) on marine environment, whole DNA, dissolved as well as particulated, was isolated from large scale sample (20 liter sea water and 10 gram of marine sediment). Using tangential filtration, ultrafiltration and resin adsorption, a proper protocol for enough DNA sample for PCR and sequencing for LMO marker was constructed. In field works as well as microcosm analysis using a bacterial plasmids (e.g. pTP1) with luminescent lux gene as a marker, vectors routinely used in many molecular biology studies were found to remain in natural marine ecosystem longer than 6 months. | - |
