Abstract
Bioassays using the embryo of bivalves have been commonly used in the quality assessment of marine environments. Although several standard protocols for developmental bioassay with bivalves have been already proposed, there was no trial to apply these protocols in Korea, or to develop a new protocol with Korean species. The South Sea Institute of Korea Ocean Research and Development Institute (KORDI) established a long-term plan to prepare the standard bioassay protocols using marine organisms commonly found along the Korean coasts. The purpose of present study is to find the optimal bioassay conditions (temperature, range of salinity, initial density of embryo) for successful development of a common mussel, Mytilus galloprovincialis, and the sensitivity of developing embryos to several chemicals (Cd, Cu, Cr, Ni, Ag, Zn). Experiments were conducted based on the standard protocol by ASTM (1989).The optimal temperature, range of salinity, and initial density of embryo were determined as 15℃, 30-35 psu, and 100 embryos/mL, respectively. At these conditions, the proportions of normally developed larvae were higher than 70%.The LOECs for Cd, Cu, Cr, Ni, Ag, and Zn were 750, 2, 188, 20, 2.5, and 8 g/L, respectively. The sensitivity of developmental bioassay with M. galloprovincialis to the metals tested was in the following order: Cu>Ag>Zn>Ni>Cr>Cd. The proportion of normal larvae appeared to decrease linearly with the logarithm of each toxicant concentration within the tested range. The average values of EC50s from 3 separate experiments for Cd, Cu, Cr, Ni, Ag, and Zn were 1797, 4.2, 786, 54.5, 6.8, and 26.6 g/L, respectively. Embryo of M. galloprovincialis showed the highest sensitivity to Cu, followed by Ag and Zn. But it showed the lowest sensitivity to Cd.
