Abstract
In the course of our screening program of bioactive marine natural products, we isolated a marine Actinomycete Streptomyces sp. from deep-sea sediments. This strain exhibited potent cytotoxic activity against human leukemia cell line(K-562). This strain was grown in modified Bennett\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\'s medium at 32C with shaking in a 5L glass flask containing modified Bennett\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\\'s medium. After 7 days, the culture broth was centrifuged and then fltrated to obtain a cell-free supernatent, followed by extraction with EtOAC. The aqueous EtOAc layer was concentrated and the residual aqueous suspension(450mg) was subject to silica vacuum flash chromatography using gradient mixtures of n-hexan and EtOAc as eluents. The fractions eluted with 40%,k and 60% EtOAc in hexane from silica vacuum flash chromatography were combined(100 mg) and purified by reversed-phase HPLC to yield a pure compound (11.5 mg) as a colorless amorphous powder. Based upon the results of combined spectral analyses, the structure of this compound was determined to be a new compound. This compound exhibited significant cytotoxic activity against human leukemia cell line (K-562).
