Abstract
In this study, we developed a thermoplastic microdevice which could handle two functions – DNA extraction and polymerase chain reaction (PCR) – in one monolith operated by a continuous flow. In detail, Cochlodinium polykrikoides, the major cause of red algal bloom in south sea of Korea, were first captured on amine-functionalized microbeads and then lysed to release DNA into subsequent PCR channel. To illustrate cell adsorption onto the amine-functionalized microbeads, Escherichia coli bacterium was used as a model system because both the E. coli and the algal cell possess negative charges on their surfaces. A 244-bp DNA target was successfully amplified from C.polykrikoides genome using a polycarbonate (PC) microdevice following the on-chip extraction, confirming the potential use of the PC monolithic microdevice for on-site analysis as well as early monitoring of the advent of red algal bloom., the major cause of red algal bloom in south sea of Korea, were first captured on amine-functionalized microbeads and then lysed to release DNA into subsequent PCR channel.
