DEVELOPMENT AND CHARACTERIZATION OF NOVEL TETRA-, TRI-NUCLEOTIDE MICROSATELLITE MARKERS FOR GIANT GROUPER (Epinephelus lanceolatus)

dc.description.abstract We isolated microsatellite loci through pyrosequencing of a repeat-enriched E. lanceolatus genomic library. A total of 143,360 raw reads were obtained from a 1/8 plate run on 454 GS-FLX plus platforms and 127,631 unique sequences were generated with an average length of 811 bp 44,578 (34.9%) sequences contained microsatellite repeats. Of the 68 loci screened, 54 were amplified successfully and 24 (8 tri-, 16 tetra-nucleotide repeat) were polymorphic with shows clear peaks in easy genotyping condition. The genetic diversity in E. lanceolatus was moderate, with an average of 6.71 alleles per locus, and the mean observed and expected heterozygosity from 0.653 and 0.633, respectively. The information generated in this study could be used for further genetic study in giant grouper such as investigate the population structure, parentage analysis, kinship selection, and so forth.ated with an average length of 811 bp 44,578 (34.9%) sequences contained microsatellite repeats.
