Abstract
Antidepressant drugs have been used to treat major symptoms of depression. The drugs may find their way to aquatic environment. To investigate influence of antidepressant drugs in aquatic organisms, we exposed O. javanicus to Fluoxetine, a common prescribed antidepressant drug. Acute toxicity determination was performed to evaluate toxicity of Fluoxetine in O. javanicus after 24, 48 and 72 h of exposure. The analysis revealed that toxicity of Fluoxetine was influenced by concentration and time of exposure. Transcript gene changes analysis was also performed to O. javanicus at environmentally relevant concentration for 12, 24, 48 and 72 h. The differentially expressed genes which were affected more than 2-fold revealed significant changes in various genes involved in nutrient metabolisms, cellular defense mechanism, cellular detoxification, and ion homeostasis. Cellular responses to drug, steroid hormone and oxidative stress were also affected. In addition, Fluoxetine also induced genes associated with behavior, reproduction and embryonic development. Changes were found in the gene expression regulated by NIF/NF-kappaß signaling and tumor necrosis factor-mediated signaling pathways as well as steroid biosynthesis, and complement and coagulation cascades. Fluoxetine induced changes in gene expression of phase I and II drug metabolyzing enzymes, and metabolism of xenobiotics by cytochrome P450.
