Abstract
Vinciguerria nimbaria is a cosmopolitan mesopelagic species of lightfish, which dwells in subtropical oceans from depths ranging from 20m to 5,000m. Its opportunistic feeding behavior and tendency to diel vertical migration (DVM) have been studied to an extent exogenously, but the endogenous factors have yet been explored. To identify genes associated with DVM of V. nimbaria, specimens were collected at different depth and time from an expedition to the Philippines Sea via the research vessel ISABU. This is the first attempt to compare transcriptome expression from mesopelagic fish to identify genes particularly associated with DVM. From RNA-seq analysis, we identified four types of opsins and seven cryptochrome subfamilies, some of which (cry1, cry5) were significantly differentially expressed. Core clock genes such as per2 were significantly differentially expressed as well. Unlike our initial hypothesis, no opsin genes were significantly differentially expressed,
while numerous photoreceptor genes as well as clock genes were significantly expressed. While our findings have identified previously unknown opsin and cryptochrome genes of V. nimbaria, as well as significantly differentially expressed genes which may be associated
with vertical migration, further analysis consisting of more specimens from more frequent phases of DVM will be required to investigate rhythmicity of peak expressions to fully understand molecular mechanism of DVM.
