Abstract
◎ Background and Objectives of This Study: Skates are commercially important fish species in Korea. Especially, Raja pulchra enjoys high consumer preference and a high price compared to other species of the family Rajidae because of its better taste. Since the morphological characters distinguishing species apart are no longer available in the processed fish products, consumers might sometimes get deceived of the species and pay more than the appropriate price. In the present study, we made an attempt to identify the species of the processed skate products in the market using the DNA barcode and infer their origin based on biogeography of the species in order to examine whether the products were labelled correctly or not in terms of the species name and its origin.◎ Methods : Thirty two samples of skates asserted as R. pulchra were obtained from fish markets and restaurants in September, 2009 and seventeen samples were collected in October, 2011 for further examination. Three specimens from each sample were used for analysis of the DNA barcode, the approximately 700bp long mtDNA COI sequence. The DNA barcode region of the COI gene was amplified by polymerase chain reaction with a pair of oligonucleotide primers, RajaCOIF [5-TCA GCC ATC TTA CCT GTG GC-3] and RajaCOIR1 [5-GGG TGT CCG AAG AAT CAG AA-3]. The PCR product was purified and sequenced in both directions.◎ Results
