Abstract
This study was to relate pathways of microbial respiration to molecular hylogenetic evidence in the unvegetated (UMF) and vegetated mudflat (VMF) covered by Suaeda japonica in Korea. Concentrations of Fe2+ at VMF were 4X higher in July (3.6 mmol m-2) and 15X higher in Octover (35.2 mmol m-2) than at UMF (0.9 mmol m-2 in July and 2.3 mmol m-2 in October). C mineralization in July was 3.2X higher at VMF (142.1 mmolC m-2d-1) than at UMF (45.4 mmolC m-2d-1), whereas C mineralization in October was only 1.6X higher at VMF (30.4 mmolC m-2d-1) than at the UMF (18.4 mmolC m-2d-1). The rate of sulfate reduction in July was 2.9X higher at VMF (53.0 mmolS m-2d-1) than at UMF (18.4 mmolS m-2d-1), but little difference was observed between VMF (9.3 mmolS m-2d-1) and UMF (10.9 mmolS m-2d-1) in October. Highest rates of C respiration and sulfate reduction at VMF in July indicated that actively growing plant roots and rhizomes excrete DOM, fueling bacterial metabolism. Molecular phylogenetic analysis of delta-Proteobacteria clones based on 16S rDNA sequence revealed that Desulfuromonas/Pelobacter using Fe(III) and So as an electron acceptor were dominating at the rhizosphere of VMF, whereas most sulfate reducing bacterial clones (Desulfobacterium & Desulfobulbus) were observed at both UMF and VMF sites. Combinations of biogeochemical measurements and molecular phylogenetic evidence indicated that, although sulfate reduction was a prevailing C mineralization pathway in the organic rich intertidal mudflat, microbial Fe(III) reduction was a significant C mineralization pathway at VMF site.
