dc.description.abstract Cyanobacteria (blue-green algae) is a phylum of bacteria that obtain their energy through photosynthesis. Aquatic cyanobacteria are probably best known for the extensive and highly visible blooms that can form in both freshwater and the marine environment.We isolated microalgae from Mangrove mud of Chuuk state in Micronesia. THe algae cultured at 30℃ in F2 medium. The species was growed between 5 to 35 ‰ Salinity and pH 7 to 9.5. Genomic DNA was isolated by normal bacteria genomic DNA isolation method. We designed a pair of primer by compare with 16s ribosomal DNA of other cyanobacteria species. PCR was perfomed for amplification of 16s rDNA with forward (GCGKCHGATTAGCTAGTTGG) and reverse primer (CGDATTCACCGCMGTATGC). Amplified gene was clonedinto pGEM easy T vector and transformed into E. coli DH5alpha. Nucleotide sequence was analyzed by Solgent co. Analyzed sequence was showed 1123 base pairs. We screened similar sequence by nucleotide Blast program from NCBI database. To determine the relationship with other cyanobacteria spicies, a phylogenetic tree was constructed based on 16s rDNA nucleotide sequence using the Neighbor-joining method in MEGA 4.1. The strain was showed 97.3% similar to 16s rDNA sequence of Dermocarpella incrassate SAG 29.84 [Accession number : AJ344559]. -
