dc.description.abstract Recently, we reported the genomic analysis of the hyperthermophilic archaeon T. onnurineus NA1 showing the presence of multiple hydrogenase gene clusters in the genome. Analysis of the hydrogenase gene clusters revealed the presence of dehydrogenase genes that were associated with the hydrogenase gene clusters, such as carbon monoxide dehydrogenase (Codh) and two formate dehydrogenases. In addition, proteins that are involved in soluble starch hydrolysis, and proteins in the EMP central pathway, were also found in T. onnurineus NA1. These data indicate that T. onnurineus NA1 may be able to grow with carbon monoxide (CO), formate or starch as a carbon source. In this study, we have experimentally investigated the cell growth and hydrogen production by T. onnurineus NA1 in media supplemented with CO, formate, or starch. From a series of batch cultures in 150 mL serum bottle, it was turned out that T. onnurineus NA1 could grow anaerobically by producing hydrogen utilizing CO, formate, or starch even though showing some differences in the rate of cell growth and hydrogen production depending on the substrate. This microbe is the first example that a single microorganism can grow and produce hydrogen using CO, formate, or starch as the sole carbon source. -
