Abstract
Sebastes koreanus, an endemic Korean species, is distributed in the Yellow Sea. We analyzed the mitochondrial DNA of S. koreanus to genetically characterize the species. Specifically, we sequenced the mitochondrial DNA control region (D-loop), cytochrome c oxidase subunit 1 (COI) gene, and cytochrome b gene (Cytb), conducted a homology search, and analyzed nucleotide variations. The D-loop region of S. koreanus was composed of 791 nucleotide sequences. Among them, 20 nucleotides (2.53%) showed variations, mostly nucleotide transitions, and 14 individuals had nine haplotypes. We analyzed a 1,075-bp fragment of the COI gene and found that the COI gene of S. koreanus was similar to that of Sebastes schlegelii. Although the C nt722 of S. koreanus was replaced by a T in S. schlegelii, the amino acid sequences did not differ. The mitochondrial Cytb sequences of S. koreanus differed from those previously reported (GenBank Accession No. AB454090), and amino acid sequence variation was detected. Further investigations were conducted using the obtained sequence information. Additionally, PCR-RFLP was performed using two specific restriction enzymes, AluI and HincII, which were able to identify S. koreanus precisely and quickly.
