Abstract
Evaluation of species occurrence and recruitment is an important step toward biodiversity assessment and is a necessary procedure in the studies of biogeography, conservation biology and ecology. Monitoring of the distribution and recruitment through identification of the larvae becomes important for the establishment of proper resource management and conservation plans.On the other hand, it is often difficult to identify plankton species due to their small size and indistinguishable morphological characters. Here we present a molecular approach based on the DNA barcode to detect presence of a target species in the multi-species mixed samples. We developed species-specific primers and probes for marine invertebrates common in the Korean waters which can amplify mitochondrial DNA sequence in the 5 region of the COI gene and distinguish the presence of each species in the netted samples. The horizontal and vertical sampling using 200um and 60um net were carried out from the Tongyung waters twice a month in 2013. Through the realtime-PCR with the species specific primer sets, the presence of the target species such as Paracalanus parvus, Calanus sinicus and other invertebrates in the mixed net samples was examined and also verified by the result of next generation sequencing.Enabling species identification from the mixed net samples, this molecular technique using the DNA barcode opens a new perspective for thent through identification of the larvae becomes important for the establishment of proper resource management and conservation plans.On the other hand, it is often difficult to identify plankton species due to their small size and indistinguishable morphological characters.
