Abstract
Ieodo and Gageocho are located in a crossroad of Taiwan warm current coming into the Korean waters. Furthermore these regions are known as one of the places with high species diversity. Zooplankton constitutes an important component of marine ecosystems, providing a link between phytoplankton production and higher trophic levels. In this study, we analyzed the biodiversity and community structure of zooplankton in the four regions in the Southen coastal waters of Korea (Ieodo, Gageocho, Jejudo and Tongyeong) using DNA metabarcoding. Zooplankton samples were collected in June 2013 and July 2014 by 60um and 200um plankton nets. DNA was extracted from the samples and the short COI barcode region (313bp) were analysed by metagenetic analysis using the illumina MiSeq sequencing platform. A total of 143 OTUs (32 OTUs from Ieodo samples, 37 OTUs from Gageocho, 53 OTUs from Jejudo, and 76 OTUs from Tongyeong) were identified to the species level as they showed more than 97% sequence similarity with GENBANK or lab-owned DNA barcode sequences. The results show that the zooplankton biodiversity is the highest in Tongyeong and that some species such as Paracalanus parvus, Oithina similis, Euphausia apacifica, Euphausia concinna, and Themisto gaudichaudii appear in all the regions. Sorensen-Dice Similarity index (SSD) estimating the similarity of species composition between the regions shows that the zooplankton community structune ecosystems, providing a link between phytoplankton production and higher trophic levels.
