Abstract
To conserve the marine resources and investigate the biological responses to the environmental stresses in Saxidomus purpuratus[1], the molecular approaches using genes and gene expression changes were attempted. RNA was extracted from the clam (1– 7 cm) and RNAseq proceeded using NGS (Next Generation Sequencing) method. The oligo microarray was constructed based on the gene sequences from the assembled nucleotides sequence results in NGS of Saxidomus purpuratus. To find out the differentially expressed genes against the environmental stresses like as seawater temperature, salinity and dissolved oxygen, we exposed the clam to those stressors for 24 or 48 hrs. As results, we found significant gene expression changes in several functions of gene category and mainly those genes belonged to signal transduction and energy production and conversion. The oligo microarray of Saxidomus purpuratus would be used for exploring the differentially expressed genes (DEG) after exposure to the stress factors and the gene functions as well as transcriptional changes of DEG will be further studied for the effects of the stressors on Saxidomus purpuratus at the molecular level.clam (1– 7 cm) and RNAseq proceeded using NGS (Next Generation Sequencing) method.
